Background Streptavidin is a tetrameric protein secreted by Streptomyces avidinii which binds firmly to biotin. Streptavidin is widely used in molecular biology through its unique high affinity for the vitamin biotin. The dissociation constant (Kd) of the biotin-streptavidin complex is about ~10-15 mol/L. The strong affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits. The streptavidin/biotin system has one of the biggest free energies of association of yet observed for noncovalent binding of a protein and small ligand in aqueous solution (K_assoc = 10**14). The complexes are also extremely stable over a wide range of temperature and pH.
Description Horseradish Peroxidase Conjugated Streptavidin is manufactured by covalently conjugating streptavidin and horseradish peroxidase (HRP) to a hydrophilic polymer backbone. This conjugate is useful for highly sensitive detection of biotinylated proteins, DNA, RNA, etc. The HRP-Streptavidin-biotin staining and detection strategy is used when increased assay sensitivity is desired for a wide variety of systems. Streptavidin is a 60 kDa tetrameric protein capable of binding up to 4 biotin molecules. Unlike egg-white avidin, streptavidin is nonglycosylated and is essentially neutral in charge, whereas avidin is basic at neutral pH. Upon this property, streptavidin has considerably less non_specific binding resulting in less background. Horseradish peroxidase is a 40 kDa glycoprotein containing ferrous heme and produces a coloured, fluorimetric, or luminescent derivative of the labeled molecule when incubated with a proper substrate, allowing it to be detected and quantified.
